I’ve been trying to find some warm fingerless gloves because of #Raynauds and basically everything REI has seems to be terrible. These were linked on raynauds dot org and I can’t buy them because I have died of wholesome. Inventor and patent holder at 12! Ran the business with her mom! Well, of course posties can get #Raynauds disease too and may have to come in and lie on the floor clenching their teeth and vocal chords while they warm up while feeling returns and nausea subsides at this time of year so they’re safe to drive again.  What do you actually expect?  Trouble is, now their movements are being tracked by GPS.

It's also amusing to see how many co-morbidities there are between stuff I have, am suspected to have and ADHD. Like hypermobility. I might have #raynauds , low magnesium which I figured out I have before I figured out I'd ADHD. I also suffer low sodium which is also an ADHD thing. Like seriously. I'm making a list of stuff for my appointment in July and it's just like I'm listing out symptoms of it but in a really weird and chaotic fashion.

There's some good news that snuck by me last year. Someone actually looked at our problem and might have found the genes involved. I absolutely did not expect to see that in my lifetime, but I'll take it!

Current treatment options are limited and unspecific. Biological mechanisms leading to the phenotype remain uncharacterized. Using genetic and electronic health record data from the UK Biobank, the Mass-General Brigham Biobank, the Estonian Biobank, and the FinnGen study, we identified 11,605 individuals with a diagnosis for Raynaud's syndrome and 1,116,172 population controls. We found eight loci including endothelial nitric oxide synthase (NOS3), HLA, and a notable association at the alpha-2A-adrenergic receptor (ADRA2A) locus (rs7090046, P = 3.93x10-47), implicating adrenergic signaling as a major risk factor with Raynaud's syndrome. We further investigate the role of the variants and ADRA2A expression in functional and physiological models. In silico follow-up analysis revealed an expression quantitative trait locus (eQTL) that co-localized and increased ADRA2A gene expression in tissue specific manner in the distal arteries. Staining with RNA scope further clarified the specificity of ADRA2A expression in small vessels. We show by CRISPR gene editing that the SNP region modifies ADRA2A gene expression in pulmonary artery smooth muscle cells. Finally, we performed a functional contraction assay on smooth muscle cells in cold conditions and showed lower contraction in ADRA2A-deficient and higher contraction in ADRA2A-overexpressing smooth muscle cells. Our results indicate that Raynaud's syndrome is related to vascular function mediated by adrenergic signaling through ADRA2A. Question for #disabled/chronically ill Mastodon folk with conditions that cause issues with cold e.g. #dysautonomia, #raynauds ... I've been doing more swimming and aquafit (good for me in many ways!) but I'm seriously struggling with cold recently. Warming up after swimming is taking *hours*. Are those long-sleeved swimming costumes any good? Anyone found any other solutions? #disability [ETA: I swim in an indoor pool, not wild swimming. My cold intolerance is pretty bad!]
